I'm in the process of building a live/drum room that is completely decoupled from surrounding walls and ceiling. The duct work is finished with drywall around it, so I was going to frame up a wall, and butt it up under the duct work flush with one side (see pic below). The inside wall of my live/drum room would then be spaced off of that about 1", filled with insulation and 5/8" dry wall.

But, I got to thinking. Sound from inside the room would basically be going through the 5/8" drywall, insulation, 1" gap, then directly into the drywall and into the duct work. Is this going to provide enough isolation for drums not to travel throughout the house, or should I frame both walls up on one side of the duct work making my drum/live room another 4" smaller?

How much better isolation would the "Potentially Better Plan" have over the original, is it worth the loss in space?

Plan "A" ("potentially better") is the way to go, for sure, but do try to leave a small gap between the drywall on the studs and the duct. Even a thin gap there would be good, to keep structure-borne vibrations separated.
